Accessioning Lab

Acceptance

Tests arrive by mail around 4:30-5PM EST

First orders are opened up (for these boxes, they are cut via exacto knife on the crease of the box)

Visual inspection of the order to confirm that there is:

Liquid in the vial

There is no damage

The swab is in the vial

Note: These are accessioned in groups of 95 and put onto racks

First QC step of a 2nd Accessioing step is made to confirm that these number work

Plating

Samples are put on a plate based on what the computer tell it to do.

Note: There is a control that is randomly assigned

QC second verification of the samples. During this step the caps are removed (and any debris removed as well).

The the samples are put onto a metal rack that enables a automate machine to pipet samples into a plastic tray

Extraction Lab

Extraction

SAmples are then moved from the Accessining lab to the Extraction Lab

They use a i7 beckman coulter machine to extrac DNA from 20 plates at a time

it takes 5 hours

Uses a bead based extration

Moved the plates to a new plate

Quantification

Samples are moved to the i5 machine where they undergo Picogreen to measure the amount of DNA in each sample.

At this step any sample that does not have enough DNA is removed

I THIKN: This is also where samples are leveled off at with th same dilution or concentration

Array Preparation

Samples are put on a EPIC chip

Epic ship is heat hybridized and cleaned off

Epic chip is then run on array machine
